Viral Vector Facility

Equipment

The Viral Vector Facility owns shRNA, CRISPR (gRNA) and ORF (cDNA) libraries from Sigma Aldrich. Constructs from these libraries are arrayed as single clones into 96-well plates and stored as bacterial glycerol stocks at -80°C freezers. The Viral Vector Facility uses the ML-II virus labs at the LUMC department of Cell and Chemical Biology (CCB).

ML-II virus labs and shRNA/CRISPR/ORF libraries

We have the following equipment  for production of viruses:

  • Biosafety level ML-II labs to produce and transduce adenoviruses, lentiviruses and reoviruses.
  • Beckman Coulter Optima XE-90 ultracentrifuge, to concentrate and purify viruses.
    Available rotors: Sw32 Ti, Sw41 Ti, 70.1 Ti and more.
  • Leica DMi8 fluorescence microscope, to monitor cells and expression of fluorescent genes.
    Available filters: GFP, CFP, DsRed, DAPI, TXR.
  • BioRad iMark microplate reader. Used for p24 Elisa to measure titers of lentiviral batches.

We offer constructs from our shRNA, CRISPR (gRNA) and ORF (cDNA) libraries:

  • Human and mouse Mission shRNA libraries of Sigma-Aldrich and The RNAi Consortium (TRC). Complete TRC1 and TRC1.5 libraries are available and the main part of TRC2 (all validated shRNA constructs, and all constructs of target genes which are not already present in TRC1 or TRC1.5).
