Viral Vector Facility
We obtained shRNA and CRISPR libraries for studying gene knockdown and gene knock-out respectively. These shRNA and CRISPR constructs can be used to produce lentiviruses.
Services
You can ask us to produce lentiviruses, adenoviruses and reoviruses for you. We can concentrate and purify viruses these viruses, and determine titers of these batches. We also can assists in producing viruses by yourself and perform infection/transduction experiments with these viruses.
You can request bacterial cultures of clones from our arrayed shRNA, CRISPR (gRNA) and ORF (cDNA) libraries from Sigma-Aldrich (Merck), for use within LUMC.
Equipment
- Biosafety ML-II level labs: for working with lentiviruses, adenoviruses, reoviruses and adeno-associated viruses
- Beckman Coulter Optima XE-90 ultracentrifuge and rotors: to concentrate and purify viruses
- Leica DMi8 fluorescence microscope: to monitor expression of fluorescent genes.
- Eppendorf centrifuge 5810R, temperature controlled, rotors for tubes and multiwell plates: used for general tissue culture and spin transduction
- Enzo Byonoy microplate reader: used for p24 ELISA to measure titers of lentiviral batches.
- Sigma-Aldrich (Merck) arrayed shRNA, CRISPR (gRNA) and ORF (cDNA) libraries: to knockdown/knock-out/overexpress genes.

We operate under a hybrid funding model, with institutional support and user fees covering operational costs.
About us
Our aim is to provide researchers access to state-of-the-art viral-vector technology of high and reproducible quality, allowing them to generate sufficient data to apply for external grants.
The Viral Vector Facility is embedded within the department of Cell and Chemical Biology (CCB) at the LUMC.
