Protein Facility

Equipment

The LUMC protein facility is equipped with a range of advanced instrumentation supporting the full workflow of protein production, purification, and analysis. It includes systems for recombinant protein expression, chromatography-based purification, and biophysical and functional assessment tools to evaluate protein quality, stability, and activity. Complementary analytical equipment enables accurate measurement of protein interactions and structural integrity.

Together, these resources provide a robust and efficient platform for producing high-quality proteins and supporting a wide range of research applications.  

Protein expression systems

  • 6 Multitron (Infors-HT) shakers: for temperature-controlled bacterial growth and expression of proteins (up to 10L per incubator)
  • 1 Large scale centrifuge: for efficient cell harvesting
  • 2 Innova shakers: for insect cell culturing (up to 8L)

Protein purification

  • 3 Bio-Rad NGC FPLC : chromatography platforms for protein purification, including affinity, ion exchange, and size exclusion chromatography
  • 1 Bio-Rad NGC FPLC Mini: chromatography setup for analytical size exclusion chromatography

Biophysical characterization

  • Multiple Bio-rad and Invitrogen electrophoresis setup: for assessment of protein purity and integrity, including native and denaturing gel-based analyses
  • Tycho NT 6.1: for rapid evaluation of protein stability and thermal unfolding behavior
  • Wyatt MALS: for accurate determination of molecular weight and oligomeric state in solution
  • Wyatt Nanostar: for small-angle light scattering for structural and conformational analysis in solution
  • Unchained labs Aunty: for high-throughput screening for protein stability, formulation, and aggregation behavior

Molecular interactions

  • BMG LabTech ClarioStar: high-performance multimode microplate reader (absorbance, fluorescence intensity, luminescence, fluorescence polarization, and time-resolved fluorescence )
  • TA Instruments Affinity ITC: for measurement of biomolecular interactions directly in solution by detecting heat changes during binding events
  • Cytiva Biacore T200: surface plasmon resonance (SPR) platform for real-time biomolecular interactions measurements without labeling (kinetics (association/dissociation rates), affinity (KD), and binding specificity)
  • Delta Life Sciences InQuiQ: for label-free biomolecular interactions measurements in real time
