LUMC Peptide Facility
Equipment
We use modern peptide synthesis, purification, and analysis equipment to support both routine and advanced projects. Our setup combines parallel peptide synthesis with strong quality control and purification capabilities. This allows us to handle standard work as well as complex peptide designs.
Equipment overview
Our facility routinely uses Fmoc solid-phase peptide synthesis and has five Syro robotic peptide synthesizers. For analysis and purification, we use WATERS Acquity H-Class UPLC systems, LCT-Premier, RDa, QDa and XEVO G2-S QTOF mass spectrometers, WATERS HPLC systems with UV-VIS detection and mass-triggered collection, and a Shimadzu LC-20 system for small-scale purifications.
Peptide synthesizers
CCB-Peptide Facility:
- 2x Syro II robotic synthesizer: parallel synthesis system for multiple peptides. These systems can synthesize 48 peptides at 25 µmol scale.
- Syro II robotic synthesizer: high-throughput parallel synthesis system. One system can produce up to 576 peptides in a single run at 4 µmol scale.
IMMO-Peptide Facility:
- Syro I robotic synthesizer: typically synthesizes 24 peptides simultaneously at a 10 µmol scale and at elevated temperatures.
- Syro II synthesizer supports parallel peptide synthesis, the system typically synthesizes 96 peptides simultaneously at a 10 µmol scale
Analytical instruments
- WATERS Acquity H-Class UPLC with Waters ACQUITY Tunable UV/Visible (TUV) and Detector Waters RDa Mass Spectrometer: used for LC-MS analysis quality control.
- WATERS Acquity H-Class UPLC with XEVO G2-S QTOF Mass Spectrometer: used for high-resolution mass analysis.
- WATERS Acquity H-Class UPLC: used for peptide purity analysis.
- Waters ACQUITY UPLC system with Waters ACQUITY Premier Tunable UV/Visible (TUV) Detector and ACQUITY QDa II Mass Detector used for LC-MS analysis quality control.
- Microflex Bruker MALDI-TOF mass spectrometer.
Purification systems
- WATERS 2545 LC with 3100 Mass Spectrometer: used for mass-triggered collection during HPLC purification.
- WATERS 2535 with 2545 UV-VIS detector: routinely used for large-scale purification of ubiquitin(-like) proteins.
- Shimadzu LC-20 system: used for small-scale peptide purifications of 1–5 mg.
