1. Isolation of primary cells:

• skin fibroblasts from punch biopsy: €436,-
• PBMCs from peripheral blood: €334,-
• cells from urine samples: upon request

2. Reprogramming/ hiPSC generation per donor sample includes:

• mycoplasma testing
• expansion of erythroblasts from PBMCs (if applicable)
• reprogramming, picking, expansion and cryopreservation of up to 6 hiPSC clones
• testing for absence of SeV (if applicable; characterized clones only)
• a maximum of two reprogramming rounds

Reprogramming skin fibroblasts with:

• Sendai virus: €4762,-
• srRNA: €4457,-
• episomal plasmids: €3780,-

Reprogramming erythroblasts (from PBMCs) with:

• Sendai virus: €4246,-

3. Genetic modification of hiPSCs with CRISPR/Cas9:

• generation of small scale knock-in, knockout or reporter hiPSCs using a ssODN
• sgRNA and ssODN design
• hiPSC electroporation with a maximum of 2 sgRNAs
• single cell cloning
• screening by PCR/ restriction enzyme analysis and sanger sequencing
• expansion and cryopreservation of up to 3 successfully edited clones

Note that off-target analysis, functional validation of knockout hiPSC or reporter hiPSC is not included in our service.

4. Standard characterization/QC for newly generated hiPSC lines or genetically modified hiPSCs, per clone:

• Pluripotency status/FACS: €81,-
• Functional pluripotency/ trilineage differentiation and IF staining: €174,-
• Cell identity/ STR analysis: €42,-
• Genetic integrity (iCS digital ddPCR): €175,-

5. Provision of extensively characterized ‘control’ hiPSC lines or hiPSC lines with STRAIGHT-IN landing pads:

Provided at no costs.

6. Participation in the annual training course with Stem Cell Technolgies:

No costs apply. The number of LUMC participants is limited.
