Leiden hiPSC Centre

Equipment

Our facility owns two state-of-the-art electroporation devices and features a single cell dispenser and imager which is especially suitable for hiPSC cloning. We also own a system for automated nucleic acid isolation and a device for capillary electrophoresis analysis of PCR products. In 2027 we plan to install an automated system for hiPSC culture including a liquid handler, a robotic arm and incubators.

Our equipment

  • Leica M80/M60 Stereo Microscope built into the LAF cabinet and EVOS M5000 Imaging System (Invitrogen): Used for QC of cultured undifferentiated hiPSCs and manual removal of spontaneous differentiation
  • Neon NxT electroporator (Invitrogen), single and 8 channel: For electroporation of hiPSCs with CRISPR/Cas9 for genetic modification
  • Nucleofector 4D, X-unit (Lonza): for reprogramming primary cells with episomal plasmids
  • SIGHT single cell dispenser and imager (Cytena): Allows efficient single cell cloning hiPSCs in the 96 or 384 well plate format. Cellular outgrowth can be monitored with the built-in imager.
  • QIAxcel Connect Capillary Electrophoresis instrument (QIAGEN): capillary-based system for analysis of PCR products
  • Maxwell for automated DNA/RNA extraction (Promega)
